1. Label 5 microfuge tubes with the following pHs: 2, 3, 5, 7, 9. (whatever is used in class that day) 2. Using a plastic pipette, fill each tube up to the 0.5ml line with the appropriate pH buffer. 3. Add 150L of mixed lactose to each tube. 4. Close the lid of each microfuge tube and mix the contents. 5. Add 150L of lactase solution to each tube. 6. Close the lid of each and mix again. 7. Place all tubes in the 37C incubator and wait for 10 minutes. 8. Place a glucose strip in each tube for 1 second, remove it and wait for X seconds, and then use the color chart to determine the amount of glucose in the tubes in mg/dL. 9. Make a data table of glucose concentration at these different pHs. (2 pts) 10. Make a line graph of glucose concentration at these different pHs.
PH Level | [Glucose] (mg/dl) |
2 | 0 |
3 | 0 |
5 | 300 |
7 | 100 |
9 | 0 |
