A colon cancer cell line was treated by decitabine (DAC) and vitamin C (VC). The
concentration of DAC was always 1nM. There are three different concentrations of VC
(VC1 is designated as a control).
We would like to know whether the different treatments have different effects on gene
expression. In addition, we also want to know if treat the cells for a longer time would
result in more gene expression. So, we treated the cells for two different lengths of time:
12 hours and 24 hours. Each treatment had 4 biological repeats. Reactivation of an
epigenetically silenced gene was measured by qPCR. Delta CT (the larger delta CT
means the more gene expression) was reported in the table that summarized the results.
1. What kind of statistical analysis would you choose to do?
2.If you are only interested in figuring out if the 3 treatments have different effects on the
reactivation of the gene in cells being treated for 12 hours only, and you designed a post hoc
analysis in your research in case you have a statistically significant result from your ANOVA
analysis. What would be the most appropriate post hoc analysis? (1 Pt)
3.If you are only interested in figuring out if the 3 treatments resulted in different mean delta CT values in cells being treated for 24 hours only, and you designed a post hoc analysis in your research in case you have a statistically significant result from your ANOVA analysis. Perform the calculations of the ANOVA test by hand. Clearly state your H0 and alpha. Draw your conclusions. If the result is statistically significant, perform the post hoc analysis by hand. (8 Pts)
