(17 pts) We have successfully produced a new protein (MW=40,000) in soluble form using cell-free protein synthesis (CFPS). We would now like to develop a purification scheme for this protein. Propose and describe a two-step chromatography process to recover the protein from the CFPS solution. Briefly explain your selections and the relevant purification conditions (buffer requirements, binding and elution conditions, etc.) Assume that the two major macromolecules (i.e. contaminants) we are trying to separate from our protein are the bacterial elongation factor Tu (EF-Tu; pI = 5.3) and the endogenous flagellar protein Flio (pI = 10.9). The pI of our protein is around 6.8.
