Step 2 - Graph the fragment sizes and distance traveled on semi-log oraph paper. Click here if you need more help in understanding how this is done
Set up semi-log graph paper with distance migrated as the X (arithmetic) axis and log base pair (p) length as the Y (logarithmic) axis. Then plot distance migrated vs. base pair (bp) length for each Eco RI Bam IlI and Hind Ill fragments.
1 Label your graph paper with "Distance Traveled in mm** for the N axis (borizontal) and "Size of DNA Fragments in bp* for the Y'axis (vertical).
2 Label the N axis (horizontal) in mm metric units with divisions at every em. Ideally you should get 70 - 100 mm (7 - 10 cm) along the horizontal axis of your graph paper.
3 Label the Y axis (vertical) in logarithmic bp units. Indicate logarithmio divisions at the Log 10, 100, 1000, 10,000 and 100,000 bp.
4 Plot the Ladder coordinates for the six fragments (distance traveled in mm -vs- fragment size in bp) onto your graph paper.
See Ladder fragment sizes in Fig 11.6 above.
See Ladder fragment distance traveled in mm for the six fragments in Table 11.1 above.
1. Plot the Uncut coordinates for distance traveled in mm -vs- fragment size in bp) onto your graph paper.
2 Using a ruler connect all the data plots on your graph paper with a line.
3 Using a straight edge calculate your "Best Fit" line by having the straight edge bisest as close to all plots equally.
Draw your Best fit line
5 Use the Best Fit line to determine the fragment sizes for the Bam HI, Eco RI, and Double Digests.
6 Record the fragment sizes in bp in table 11.1 above.
