1. (20 points) For the determination of the enzymatic activity of alcohol oxidase (AO) from yeast, the following coupled reactions are utilized: AO methanol + O2 formaldehyde + H2O2 HP H2O2 + DH2 -> 2H2O +D where DH2 is a dye precursor, D is the dye, and HP is the enzyme horseradish peroxidase. The dye is monitored by means of a spectrophotometer at a wavelength of 405 nm. The standard curve for the assay (Figure 1) is prepared using various concentrations of H2O2 in the reaction mix with no AO or methanol added, using a light path of 10 mm. 0.7 0.6 0.5 0.4 A40s, absorbance at 405 nm y = 0.0377x -0.0123 0.3 0.2 0.1 0 0 5 15 20 10 Concentration of H2O2 (umol/liter) A sample of clarified yeast cell lysate assayed for AO activity by this method gave a rate of change of A4os of 0.075/min in the assay after a dilution of the sample of 1:30,000. Calculate the AO concentration in units per milliliter, where 1 unit (U) is equal to the amount of enzyme producing 1 umol of H2O2 per minute under the conditions of the assay
