24. A study on the capability of bacteria Enterobacter cloacae in cleaning up oil spillage was studied. The batch culture of Enterobacter cloacae was carried out at 28C. The substrate, kerosene, with an initial concentration of 10g/L was fed into the lab scale bioreactor. The inoculum of 0.1g/L was added. The cells are growing actively with a doubling time of 2 hours. The system came to the stationary phase after 11 hours of reactor operation, where the kerosene was completely consumed. (a) Estimate the maximum specific growth rate (max) of the microorganism and the yield coefficient (Yx/s). State the assumptions made. ( 8 marks) (b) Given the maximum specific growth rate of Enterobacter at 35C is 0.8h1, estimate the maximum specific growth rate of the cell at 32C. (8 marks) (c) With the answer obtained in (b), compare the time taken for cell concentration to reach 5g/L at 32C and 35C. (4 marks) (d) If 2g/L instead of 10g/L of kerosene was used initially at 28C, predict the total time needed for the culture to reach stationary phase, by assuming that SKs during the entire stage of the culture.
