Question
A researcher is preparing to perform a western blot to compare Estrogen Receptor (ERa) and Her2 protein levels in normal (N) breast and breast tumour
A researcher is preparing to perform a western blot to compare Estrogen Receptor (ERa) and Her2 protein levels in normal (N) breast and breast tumour (T) biopsy tissues from 3 patients.
She has prepared total protein extracts from equal amounts of each tissue biopsy. The concentrations of the protein extracts (in mg/ml) are shown in the Table below.
- To compare expression levels of ERa, she needs to load an equal amount of each other sample. Calculate the volume of the other samples required to complete the Table above. Give the answers in ul to 1 decimal place.
- calculate the amount of total protein (in mg) from the tumour tissue (T1) extract she has loaded on the gel.
After separating equal amounts of each extract by SDS PAGE, the gel was blotted and the researcher used antibodies to detect ERa, and HER2 proteins. As a loading control she also detected the relative levels of B-actin protein in the extracts. Based on the results obtained in the blots shown below, fill in the Table to identify which patient(s) might be suitable for treatment by antihormone or Herceptin therapy?
