You will be testing 7 different concentrations of your reagent/chemical to determine the EC50 your Qeagent/chemical. 1 Make the dilutions of the different compounds including vehicle and H2O2 control. a. Make sure to calculate the volume needed from your stock solution using the formula below. (Stock solution is at 400 uM) V1C1=V2C2V1=C1V2C2 For your TEST compound, you will test 7 different concentrations. Column 1 - 66M Column 2 - 33M Column 3 - 16.5M Column 4 - 8.25M Column 5 - 4.125M Column 6 - 2.0625M Column 7 - 1.03125M Making your series of different compound concentrations 1) Label your microcentrifuge tubes \#1 through \#7 (\#1 will be the highest concentration and \#7 will be the lowest concentration). 2) To tubes \#1 - \#7, add 250L of MEDIA (DMEM 10\%FBS). 3) To tube \#1, add 250L of the stock compound; close cap and vortex. 4) Transfer 250L from tube \#1 to tube \#2; close cap and vortex. 5) Transfer 250L from tube \#2 to tube \#3; close cap and vortex. 6) Repeat this procedure for each subsequent tube until tube \#7; for each dilution, close cap and vortex before proceeding with the next dilution. 7) Now you have your Dilution Series for your compound
