In need of help! Not sure how to find keep the concentration the same for 10X BCP and get different ratios of BCP:BSA... 10 uL of 10X BSA = 7.5 uM Need to keep concentration of BCP at 6uM Looking for the methodology here, not necessarily the answers! Maybe an example or two, but I need to understand the how behind the answers.
The ranges of the two instruments might be slightly different. Do your new results confer with your early tests of these instruments? For maximum signal, adjust the starting dye concentration (i.e. use 1.3X instead of 1x). Each sample will have a fixed concentration of BCP based on your calculation from above (i.e. all entries in column 2 should be the same, somewhere between 100 uL and 200 uL). Plan your experiment so that reaction #5 has a 1:1 stoichiometry (reactions 1 - 4 will be sub-stoichiometric).
