Day 2 A. Preparation of Affi-Gel Blue (AGB) 1. Transfer 1.00mL of uniform Affi-Gel Blue suspension to an Eppendorf tube. Before transfer, ensure that the resin is fully, uniformly resuspended. 2. Spin down in a microcentrifuge at maximum rpm for 5min. 3. Carefully remove the buffer with a Pasteur pipet, leaving 0.500mL of packed AGB in the centrifuge tube. Place tube in an ice bath. B. Batch Purification of LDH NOTE: Always use different pipet tip for each different ENZYME solution. NOTE: During incubation periods, when you have nothing else to do, feel free to do step D. 1 to. save some time later today. 1. Take 0.400mL of ammonium sulfate suspension of LDH and add 1.60mL of 0.0500M phosphate buffer, pH7.50 ( 15 dilution of AS suspension). Place in ice bath. (This sample is referred to as the B-1 solution; label it as such) 2. Transfer 1.00mL of this LDH solution to Eppendorf tube containing AGB. Vortex gently. 3. Allow mixture to stand in ice bath for 5min, vortexing gently once per min. 4. Centrifuge for 3min at maximum rpm. 5. Carefully remove the supernatant with a Pasteur pipet and discard. Note: It is better to leave a small amount of the supernatant behind with the resin than to discard any of your resin. 6. Add 1.00mL of 0.0500M phosphate buffer pH7.5 to your resin, vortex it and centrifuge for 3 min at max rpm. Remove supernatant and discard. 7. Repeat step 6. 8. Repeat step 6 once more. 9. Add 1.00mL of Specific Elution Buffer (2.00mMNADH/0.200MNaCl/0.0500M phosphate buffer, pH7.50 ) to washed AGB from step 8 and vortex. 10. Allow mixture to stand at room temperature for 15min. Vortex once per min. 11. Centrifuge for 3min at max rpm. 12. Carefully separate supernatant with a Pasteur pipet and save in a test tube in ice bath. This is your Affi-Gel Blue purified LDH solution (This sample is referred to as the B-11 solution; label it as such)
